- W199190693 abstract "ABSTRACT Environmental contaminants such as polybrominated biphenyls (PBBs) and polychlorinated biphenyls (PCBs) produce no detectable alteration in normal renal function. There is a growing body of evidence however, that these contaminants alter the biochemical pathways responsible for the metabolic activation of other chemicals which may be nephrotoxic. Chloroform nephrotoxicity was markedly enhanced by PBB ingestion in mice and the degree of enhancement was proportional to the concentration of PBBs in the diet. The nephrotoxicity of carbon tetrachloride, trichloroethylene and 1,1,2-trichloroethane was also markedly enhanced in PBB treated mice. Similarly, the nephrotoxicity of acetaminophen was enhanced in rats treated with PBBs. On the other hand, PCBs reduced the nephrotoxicity of chloroform in mice. The nephrotoxicity of chloroform was also reduced by exposure to TCDD. Thus, a variety of environmental chemicals may influence metabolism within the kidney and subsequently alter the toxicity of certain compounds that require metabolic activation to a nephrotoxic product." @default.
