- W1992371857 abstract "Development of pH glass microelectrodes (1 μm tip diam.) and a micromanipulator-illuminator system (for safe, accurate, intraretinal electrode placement) has made possible the direct in situ measurement of vitreous and intraretinal pH. Miniature pigs were anesthetized and placed on a respirator. The micro-manipulator was placed on the eye: the illuminator and pH electrode were then introduced into the vitreoas through a scleral hole. Intraocular electrode manipulation was controlled hydraulically under microscopie observation. The preretinal vitreous pH was measured under normocapnic and hypercapnic conditions. The electrode was then advanced to the inner-retina where the buffer value was determined following acute hypercapnia. The effects of: hypercapnia, metabolic alkalosis (i.v. NaHCO3), metabolic acidosis (i.v. NH4Cl) and carbonic anhydrase inhibition (i.v. acetazolamide; DiamoxR) on inner-retinal pH were tested. The following results were obtained: (1) The mean vitreous pH was 7·32, and the mean inner-retinal pH was 7·22; both were more acidic than either plasma or aqueous humor. (2) The mean calculated inner-retinal buffer value was 33·09 Slykes. (3) Hypercapnia caused a pH decrease in both the vitreous and the inner-retina, but response to CO2 administration was much slower in the vitroeus. (4) Single rapid i.v. injections of NaHCO3 and NH4Cl did not affect inner-retinal pH. (5) Acetazolamide caused an inner-retinal pH decrease similar to that of pure hypercapnia. Presumably acetazolamide did not cross the blood-retinal barrier and this decrease was the effect of hypercapnia resulting from carbonic anhydrase inhibition in the blood. Based on preretinal vitreous and inner-retinal pH behavior together with former retinal blood flow observations, it is postulated that retinal vasomotor response to CO2 is regulated by H+ activity in the glial cells, not in the vitreous (extracellular fluid). The role of the intercellular elefts (100–150 Å wide perineuronal extracellular spaces) in this response is unknown." @default.
