- W1992809093 abstract "OBJECTIVE: Cumulus cells (CCs) are considered a valuable source for non-invasively screening for oocyte competence. Several studies have shown that CC gene expression can be correlated with IVF outcome. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression post transcription. This study investigated CC miRNA profiles as a potential regulator of CC gene expression in relation to embryo potential. DESIGN: Experimental Study. MATERIALS AND METHODS: CCs from individual cumulus oocyte complexes (COCs) were collected with patient consent (n=6). Oocytes were cultured individually with embryo assessment performed on D3 and D5. Total RNA was isolated from single oocyte derived CCs (n=12) and hybridized to NCode Human miRNA Microarray (Invitrogen) or Codelink Human Whole Genome Bioarray (Applied Microarray). Data was analyzed by Genespring software (Agilent) according to the following groups: A) COCs that resulted in cleavage stage arrest embryos (n=6) B) COCs that resulted in high quality D5 blastocysts (n=6). RESULTS: A miRNA profile of single oocyte derived CCs revealed 70 known miRNAs. Eight miRNAs observed in CCs have previously been reported during human ovarian steriodogenesis: hsa-let-7b, hsa-miR-29a, hsa-miR-126, hsa-miR-141, hsa-miR-136, hsa-miR-7, hsa-miR-187 & hsa-miR-34a. Statistical analysis comparing the CC miRNA profiles of resulting cleavage stage arrest embryos with the profiles of resulting high quality D5 blastocysts revealed no significantly different miRNA gene expression. In agreement with this observation further investigation of the genes responsible for miRNA processing including mRNA for DICER and MOV10, revealed no significant gene expression differences in CC samples associated with embryo development. CONCLUSIONS: This study has demonstrated the stability of CC miRNA in correlation with subsequent embryo potential. Further investigation into other gene regulation methods responsible for CC gene expression differences relative to outcome, such as DNA-RNA transcription is warranted." @default.
