- W1992969755 abstract "Background The aim of this study was to use molecular and serological methods to investigate the prevalence of human herpesvirus 8 (HHV-8) both in patients with lymphoproliferative diseases and in healthy population from Northern Italy(the Po valley region), one of the few areas of the world where classic KS is most prevalent. Methods We used PCR to look for the presence of HHV-8 sequences in the pathologic lymph nodes of 137 cases of benign, atypical and malignant lymphoproliferative disorders (38 HIV positive and 99 HIV negative). We also used an immunofluorescence assay (IFA) to look for the presence of antibodies to HHV-8 in the sera of 47 healthy subjects, 20 patients with HIV negative lymphoproliferative diseases and 15 clinical attenders from Zaire. Results We identified HHV-8 DNA in the pathologic tissues of 3 of 17 cases of angioimmunoblastic lymphadenopathy (AILD), in 2 of 17 cases of Castleman's disease (CD), in the primary cerebral B cell lymphoma of a patient treated with long term steroids for uveitis, as well as in the lung tissue of a patient with interstitial pneumonitis and an EBV associated B cell lymphoproliferation. HHV-8 DNA was also found in 4 cases of benign localized lymphadenopathy with giant germinal center hyperplasia and increased vascularity which may represent the clinico-pathologic entity occurring during HHV-8 primary infection in apparently normal subjects. Relevant to this, in 1 of these 2 cases available for serologic analysis, IFA assay tested positive. Conclusions In our geographical area HHV-8 seroprevalence is likely to be higher than that in USA and U.K., similarly to what has been observed for Greek population with a similarly higher incidence of classic KS. Serologic analysis on a larger series of patients is undergoing to identify subsets of lymphoproliferative diseases possibly associated with a higher prevalence of HHV-8 infection. In our geographical area the presence of HHV-8 DNA is not restricted to patients with KS or AIDS, although it is mainly associated with lymphoproliferative conditions characterized by immune impairment (AILD, CD, iatrogenic immune suppression). We also report the occurrence of few cases of benign disease (localized form of benign lymphadenopathy with peculiar histologic features), associated with a serologically confirmed HHV-8 infection." @default.
