A system to demonstrate that RNA can transmit information between two cell lines for the synthesis of an enzyme should preferably lack that enzyme in the acceptor cell line. The mouse fibroblast line, ML (TK−), which is deficient in thymidine kinase does not meet this requirement since it was found by the use of heavily tritiated thymidine to contain 1 % of the activity of the parent line. 2. 2. Chromosome analysis showed that the LM (TK−) line had, as reported previously, 5 chromosomes less than the parent line but had undergone a change in a marker chromosome. 3. 3. From 6 experiments no significant increase in thymidine kinase level could be detected in the LM (TK−) cells treated with RNA from the LM (TK+) cells. 4. 4. Thymidine kinase was also assayed radioautographically by the incorporation of [3H]thymidine into cell DNA. 9 experiments were performed and showed no definite trend. In 3 of these experiments the cells were kept in growth media for 24 h after the RNA treatment and in 2 of these experiments an apparent increase in thymidine kinase level was observed. This increase was not noted in parallel experiments in which the donor RNA preparation had been treated with ribonuclease." @default.
