- W1993440171 abstract "Procedures were developed for the isolation of various subclasses of bovine serum and colostral IgG in order to facilitate the characterization of their physico-chemical properties. IgG1 and IgG2 from both serum and colostrum were isolated and purified by use of Bio-Gel and TEAE-cellulose chormatrography. Antigenic differences between colostral IgG1 and IgG2, and serum IgG1 and IgG2 were demonstrated by the use of a monospecific anti-colostral and anti-serum IgG1 respectively. Both the serum and colostral IgG1 wer found to be antigenically identical. Similarly, IgG2 isolated from both serum and colostrum were also demonstrated to possess antigenic identity.While the partial specific volume (ν) for serum IgG1, serum IgG2, colostral IgG1 and colostral IgG2 were found to be 0·7, 0·692, 0·726 and 0·692 g/cm3 respectively, the mol. wt were found to be 145,000, 146,000, 156,000 and 146,000 g/mole respectively. The extinction coefficients calculated for serum IgG1, serum IgG2, colostral IgG1 and colostral IgG2 were found to be 12·22 ± 0·17; 12·3 ± 0·06; 12·1 ± 0·1 and 12·1 ± 0·2 respectively.Serum IgG1 was found to undergo aggregation with the formation of faster sedimenting polymers in 0·01 M TrisHCl:0·32 M NaCl (0·32 ST) at pH 8·0, 6·4, 6·9 and 7·5. Aggregrates were also formed when the solvent used was 0·01 M TrisHCl:0·089 M NaCl, pH 8·0. Colostral IgG1 also underwent aggregation in observed in 0·01 M TrisHCl:0·089 M NaCl, pH 8·0. Colostral IgG1 also underwent aggregation in 0·32 ST but no aggregation was observed in 0·01 M NaH2PO4−0·14 M NaCl, pH 6·0. Similarly, IgG2 from serum and colostrum did not show any aggregation either in 0·01 M TrisHCl:0·089 M NaCl pH 8·0 or in 0·01 M NaH2PO4−0·14 M NaCl, pH 6·0 buffers. Finally, no evidence for the presence of secretory immunoglobulin G (γGs), antigenically distinct from IgG1 and IgG2, was found in our investigations." @default.
