- W1993823915 abstract "Middle-aged and older adults who engage in regular aerobic exercise demonstrate enhanced plasma fibrinolytic activity and a lower incidence of atherothrombotic events compared to their sedentary peers. However, the mechanisms by which exercise confers this thromboprotection are not completely understood. We tested the hypothesis that the capacity of the vascular endothelium to release tissue plasminogen activator (t-PA) is greater in endurance-trained compared with sedentary middle-aged and older men. As part of an ongoing study, endothelial release of t-PA was determined in response to intrabrachial infusions of bradykinin (BK: 125–500 ng/min) and sodium nitroprusside (NTP: 1–4 μg/min) in 4 endurance-trained (runners; 58 ± 2 yr; ET) and 7 sedentary (59 ± 2 yr; S) healthy men. Forearm blood flow was assessed by plethysmography. Net release of t-PA across the forearm vascular bed to each drug dose was calculated as the product of venoarterial concentration gradient and forearm plasma flow. The vasodilatory responses to BK and NTP were not different between the groups. ET demonstrated greater net release of t-PA antigen and t-PA activity across the forearm in response to BK (p < 0.01) compared with S. At the highest BK dose net release of t-PA antigen (47.7 ± 4.7 vs 30.1 ± 4.3 ng/100mL tissue/min) and t-PA activity (22.9 ± 3.1 vs 11.9 ± 2.4 IU/100mL tissue/min) were 58% and 92% higher, respectively, in the ET vs S. There was no change in the net release of t-PA antigen or t-PA activity from baseline in either group in response to NTP. These preliminary results demonstrate that endothelial release of t-PA is greater in middle-aged and older men who regularly perform endurance exercise.
