- W1994470054 abstract "Hypercholesterolemia is an early risk factor for Alzheimer's disease (AD). However, disagreement exists regarding this association. Low density lipoprotein receptors (LDLr) may be involved in the pathogenesis of AD and also in cognitive function. Our objective was to determine if cognitive abnormalities are present in patients with heterozygous familial hypercholesterolemia (FH) a condition primarily resulting from dysfunction of LDLr. We designed a case-control study to examine the odds of cognitive impairment among patients with FH relative to patients in a control group. Patients with prior history of stroke or transient ischemic attacks were excluded from the study. Neuropsychological studies were performed in 70 patients with FH older than 50 years and 70 healthy subjects matched for age, gender and education years. Cognitive performance was determined by 13 standardized neuropsychological tests. Brain imaging of patients with MCI was performed to exclude vascular or structural lesions. Compared to controls, patients with FH showed evidence of global cortical cognitive dysfunction characterized mainly by deficits in memory, visuospatial orientation and executive function. Sixteen patients from the FH group (22.9%) met criteria for mild cognitive impairment (MCI) versus only two (2.8%) from the control group (P<.001). Patients with FH showed a high prevalence of MCI. Because prior studies have shown that older patients with sporadic hypercholesterolemia do not show higher prevalence of MCI, the findings suggest that early exposure to elevated cholesterol or dysfunction of LDL receptors may be risk factors for MCI." @default.
