- W1994646598 abstract "Abstract Male rats were administered 1 or 2 g/kg ethanol by gastric intubation and sacrificed 60, 90 and 120 min later. Brain histamine, its synthesizing enzyme, histidine decarboxylase, and the metabolizing enzyme, histamine N-methyltransferase were determined. Plasma corticosterone and ethanol concentrations were also assayed. Hypothalamic and midbrain HA levels were significantly increased 120 min after ETOH (1 g/kg) while hypothalamic HD activity was significantly elevated at 60 and 90 min following drug administration. After 2 g/kg of ETOH hypothalamic HA concentration decreased significantly (90 min) while HD activity was significantly elevated in the hypotalamus (60 and 90 min), midbrain (60 min) and cortex (60 min). HMT failed to change in any brain region at any dose or time studied. These results suggest that the elevation of hypothalamic HA levels seen after the lower dose of ETOH may be due to an increased HA synthesis rather than changes in HA metabolism since HMT activity remained unchanged. HA depletion after the larger dose could be attributed to HA release and the associated increase in HD may represent a possible compensatory mechanism. Since lower doses and higher doses of ETOH have different effects on HA concentration and HD activity it is suggested that two separate dose-dependent neurochemical effects may be involved. Plasma corticosterone levels increased only at 120 min after the larger ETOH dose which suggests that non-specific stress-induced HA changes are not involved. The plasma ETOH levels attained in these studies (20–97 mg%) are clearly within the clinical range and therefore may have clinical significance." @default.
