- W1994960808 abstract "Metabolic engineering has emerged in recent years as the field concerned with the improvement of cells using modern genetic tools. It comprises a synthesis step that introduces new pathways and genetic controls implemented by molecular biological methods, as well as a step of analysis aiming at the elucidation of the properties of metabolic reaction networks in their entirety and the rigorous evaluation of the physiological state of resulting recombinant cells. The latter step of analysis relies heavily on metabolic flux determination as a fundamental metric of cell physiology and shares common concepts with chemical reaction engineering. In this sense, fluxes, along with small metabolites, mRNA transcripts, and proteins constitute four classes of intracellular variables that define the cellular physiological state in its entirety and can form the basis for modeling cellular function. Methods capable of generating data at high throughput for all four classes of variables are becoming increasingly available, raising the need for effective methods to integrate such data in comprehensive models of cellular function. Metabolic engineering provides a convenient framework for such integration. Such models will facilitate the evaluation of the impact of genetic modifications on cellular physiology and guide subsequent rounds of modifications to attain the goals of metabolic engineering." @default.
