- W1996957404 abstract "The metabolism of labelled testosterone, 5α-dihydrotestosterone and 5α-androstane-3β, 17β-diol was investigated in vitro in NADPH-supplemented or unsupplemented slices of biopsy specimens from human normal hyperplastic and untreated cancerous prostates. Respective activities of the prostatic Δ4-3-oxosteroid 5α-reductase, 17β-, 3α- and 3β-hydroxysteroid dehydrogenases and 3β-hydroxysteroid 7α-hydroxylase were thus measured. No significant difference in activity was found between normal glands and tissues with hyperplasia or well-differentiated carcinoma. In contrast, a marked decrease in the 5α-reduction of testosterone was always noticed in biopsies from poorly-differentiated carcinomas. In some cases this decrease paralleled, even in the presence of NADPH, a more extensive oxidation of testosterone into 17-oxo-metabolites. Hyperplasia or carcinoma did not affect activities of the 3α- and 3β-hydroxysteroid dehydrogenases but 7α-hydroxylation of 5α-androstane-3β, 17β-diol was more extensive in poorly-differentiated carcinoma than in normal and hyperplastic glands. Thus loss of differentiation and of cytoplasmic organelles responsible for secretion is mainly linked, in poorly-differentiated carcinomas, with a diminished formation of 5α-dihydrotestosterone, the intracellular substrate for remaining C19-steroid metabolizing enzymes and a key hormone in the expression of androgenic activity in the prostate gland. Since loss of differentiation and of testosterone 5α-reductase and increase of 17-oxidation are also effects of oestradiol on prostatic tissue, it is questionable whether oestrogen therapy affects poorly-differentiated carcinomas and whether supply of the diseased cells with 5α-dihydrotestosterone could determine the androgen-dependent cellular differentiation." @default.
