- W1997062419 abstract "The “specific heart granules” in the right atrial muscle cells of the rat were investigated by electron microscopy and cytochemistry in order to determine whether they are able to store and/or accumulate endogenous and exogenous catecholamines, respectively. For this purpose tissues from untreated and reserpinized rats were examined (a) by a specific cytochemical method, based on the chromaffin reaction, for the ultrastructural localization of endogenous biogenic amines, and (b) after either in vitro incubation of thin tissue slices in 5-hydroxydopamine-containing solutions, or perfusion of isolated whole hearts with noradrenaline-containing solutions. In neither case were there indications of the presence of endogenous or exogenous amines. The frequency, fine structure, and cytochemistry of these granules were unchanged after the various treatments. Indeed, it appeared that their electron density is related to the staining of tissue sections with uranyl acetate and not to the presence of biogenic amines. Moreover, the electron dense content of the atrial granules could be digested by treatment of the sections with pronase. From these experiments it is concluded that the specific heart granules are unable to store appreciable amounts of endogenous or exogenous amines and that their electron dense, uranophilic core consists of a pronase-sensitive, probably proteinaceous, material." @default.
