- W1997535897 abstract "Administration of amphenone or methapyrapone as 0.3% of the diet to 28-day-old female rats for 29–32 days resulted in a decrease in body weight gain, a decrease in plasma steroid concentration, and an increase in adrenal weight. Thin-layer chromatography indicated the presence of corticosterone in the plasma extracts of untreated but not of amphenone- or methapyrapone-treated rats. However, two unknown fluorescent substances detected in the plasma extracts of treated but not untreated rats may represent biologically inactive glucocorticoid precursors. Deoxycorticosterone was detected in the plasma of rats treated with methapyrapone but not of untreated rats or those treated with amphenone. Intravenous administration of corticosterone, in amounts as little as 0.001 μg, to rats in which normal adrenal steroid synthesis was inhibited by amphenone or methapyrapone resulted in an increased release of ACTH, as indicated by both a depletion in adrenal ascorbic acid and an increase in fluorescent plasma steroids. ACTH release in response to increasing doses of corticosterone followed a bell-shaped curve when plotted. The adrenal-stimulating effect of small amounts of corticoid in the drug-treated rats was unaltered by interruption of the hypothalamicohypophyseal pathways by means of median eminence lesions placed 2 days previously. These findings indicate that low levels of circulating glucocorticoids stimulate directly the cells of the anterior pituitary to secrete ACTH." @default.
