- W1997584407 abstract "Concentrations of pyridoxal-5'-phosphate in blood and urine of infants, children and adults as well as in various human tissues of fetuses and adults were determined by a sensitive radioactive method using 3H-tyrosine and micro-column chromatography. By application of a tyrosine concentration of 25 μmol/l we were able to measure PLP as low as 5 fmol/l. An important advantage of this method is that the crucial step of neutralization of the acid used for the PLP extraction can be eliminated. A multiple dilution of the mixture with the buffer of the assay (0.1 mol/l Na-acetate, pH 5.5) immediately after the denaturation step was sufficient for the analysis, hence preventing PLP destruction at acid pH. The reference values for the PLP concentration in tissues did not vary greatly, unlike those of blood and urinary PLP. Liver contained the greatest amount of PLP, most probably due to an abundance of PLP-dependent enzymes in this tissue. A relatively high level of PLP was found in brain. In blood the PLP concentration was high in infants, and subsequently decreased with age. A decrease in the urinary level of PLP with age was also observed, and was well correlated with the blood PLP concentration. PLP concentrations of blood as well as of urine were lower in the patients with neuroblastoma as compared with the age-matched controls, whereas those in biliary atresia were significantly higher. This increase of PLP levels in biliary atresia may be related to the increased activity of aminotransferase activity in blood. These results indicate that secondary cystathioninuria observed in neuroblastoma and biliary atresia is not solely due to PLP deficiency." @default.
