- W1997940106 abstract "1. The release of glycosaminoglycans from washed human platelets has been studied. The process was induced by thrombin or polystyrene latex particles in the presence or absence of extracellular Ca2+. The pattern of this release was compared to that of adenine nucleotides and β-N-acetylglucosaminidase. Glycosaminoglycans were determined as amino sugar set free by HCl hydrolysis. 2. The time course of release of glycosaminoglycans was similar to that of adenine nucleotides. Extracellular Ca2+ had no effect on the release of adenine nucleotides or glycosaminoglycans, but the release of β-N-acetylglucosaminidase was stimulated 2.4-fold in the thrombin-induced and 1.4-fold in the polystyrene latex-induced release experiments. 3. Thrombin-induced release of glycosaminoglycans was energy requiring, and inhibited by 25% after preincubation of the platelet suspension for 30 min with antimycin and 2-deoxyglucose. The corresponding inhibition values for release of adenine nucleotides and β-N-acetylglucosaminidase were 73 and 90%, respectively. 4. High voltage electrophoresis of the released compounds revealed two alcian blue-positive substances, one with the same mobility as commercial chondroitin sulphate and heparin." @default.
