Seeds within one pod differ in size, the heaviest seeds contained the largest number of cells. It depended on the initial size and number of cells, how long cotyledons could be grown in vitro on a medium without growth hormones. Cotyledons did not grow for longer than 21 days at 16°C. During that period, starch and protein were synthesized. Whole seeds in vitro showed only limited increase in mass of fresh and dry matter. Removal of the seed-coat facilitated seed growth. The initial growth rate of cotyledons was higher than on intact plants but declined after 4 days in culture. Increase in temperature caused an increase in rate of accumulation of dry matter. The optimum temperature was about 28°C. Higher temperature shortened the duration of growth and had no effect on number of cells, nor on starch and protein contents. With the medium used (without growth regulators), cotyledons could be grown in vitro for only a limited period of time. Cotyledons did not grow to maturity, because conditions were suitable only for accumulation of dry matter. Continuous production of new cells would be necessary too, in order to support the accumulation of storage products." @default.
