By this method the degradation of mono- and digalactosyl diglyceride and the formation of acylgalactosyl diglyceride in homogenized spinach leaves (Spinacia oleracea) were investigated. 3. The pH optimum for the formation of acylgalactosyl diglyceride in spinach leaf homogenates was found to be close to pH 4.6. After 3 h incubation at this pH, acylgalactosyl diglyceride accounted for about 20% of the leaf lipids. 4. A protein fraction was obtained by (N114)2804 precipitation and dialysis from homogenized spinach leaves, which catalyzed the formation of acylgalactosyi diglyceride. 5. This protein solution was used in incubation experiments with purified lipids. By quantitative determination of the galactolipids and analysis of their fatty acid composition it was found that acylgalactosyi diglyceride is formed by an acyi transfer from three different donors: (1) from monogalactosyl diglyceride as the only substrate by dismutation, (2) from digalactosyl diglyceride, when mono- and digalactosyi diglyceride were the only lipids present; in this system the dismutation of monogalactosyi diglyceride was not detected, (3) from various phospholipids; in this case, however, two-thirds of the acylgalactosyi diglyceride originated from the dismutation of monogalactosyl diglyceride. 6. It is concluded that in spinach leaf homogenates acylgalactosyl diglyceride is formed mainly by acyl transfer from di- to monogalactosyl diglyceride. 1. Es wird eine Methode zur quantitativen Bestimmung von Mono-, Di- und Acylgalaktosyldiglycerid nach dünnschichtchromatographischer Trennung beschrieben. 2. Mit dieser Methode wird der Abbau von Mono-und Digalaktosyldiglycerid und die Bildung von Acylgalaktosyldiglycerid in Homogenaten von Spinatblättern (Spinacia oleracea) untersucht. 3. Das pH-Optimum der Bildung von Acylgalaktosyldiglycerid in Homogenaten von Spinatblättern lag bei pH 4.6. Nach 3 h Inkubation bei diesem pH-Wert machte das Acylgalaktosyldiglycerid etwa 20% der Blattlipide aus. 4. Aus Spinatblättern wurde durch Ammonsulfatfällung und Dialyse eine Eiweissfraktion erhalten, die die Bildung von Acylgalaktosyldiglycerid katalysierte. 5. Diese Eiweisslösung wurde zu Inkubationsversuchen mit gereinigten Lipiden benutzt. Auf Grund der quantitativen Bestimmung der Galaktolipide und der Analysen ihrer Fettsäurezusammensetzung wurde die Bildung von Acylgalaktosyldiglycerid auf einen Acyltransfer von drei verschiedenen Donatoren zurückgeführt: (1) auf die Dismutation des Monogalaktosyldiglycerids als einzigem Substrat, (2) auf den Acyltransfer vom Digalaktosyldiglycerid, wenn Mono- mit Digalaktosyldiglycerid inkubiert wurde; in diesem Fall wurde die Dismutation des Monogalaktosyldiglycerids nicht nachgewiesen, (3) auf den Acyltransfer von Phosphatiden, wobei allerdings zwei Drittel des Acylgalaktosyldiglycerids durch Dismutation des Monogalaktosyldiglycerids entstanden. 6. Die Bildung des Acylgalaktosyldiglycerids in Blatthomogenaten wird auf den Acyltransfer von Di- auf Monogalaktosyldiglycerid zurückgeführt." @default.
