Á l'aide de l'exemple de la co-exposition aux oxydes de fer et aux hydrocarbures polycycliques aromatiques, observée en milieu professionnel et en environnement général, cet article se propose de présenter la démarche scientifique appliquée en vue d'élucider les mécanismes physiopathologiques sous-jacents potentiellement impliqués clans la toxicite additive, voire synergique, de cette exposition concomitante. Air pollutants arising from traffic clustering and industrial production include a great number of chemical compounds that at high doses play major roles in causing many forms of respiratory disease in animal models and in some instances also in humans. However, occupational and environmental exposures are generally to low doses of complex mixtures of pollutants. Direct epidemiological evidences for adverse effects of air pollution in humans is, also, weak, and most of the available studies are limited by lack of adequate control of confounding factors and other methodological drawbacks. Moreover, it is now important to consider the potential occurrence of an addition and even a synergy between the adverse effects induced by a concurrent exposure. Perspectives and approaches to a better knowledge of the involved risk factors may include not only epidemiologic studies, to evaluate patterns of health and disease in groups of persons, but also experiments to elucidate the underlying mechanisms by which pollutants induce an additive and even a synergistic toxicity. Giving the example of the occupational andlor environmental exposure to iron oxides and polycyclic aromatic hydrocarbons, this article describes such a scientific approach." @default.
