- W2000006640 abstract "Interleukin 6 (IL-6) has been related to beta-amyloid aggregation and the appearance of hyperphosphorylated tau in AD brain. However, previous studies only explored one or two single nucleotide polymorphisms (SNPs), which were unable to capture the majority of genetic information of IL-6, and results were inconsistent. This is a case-control study. A total of 284 Alzheimer's disease (AD) patients aged 50 or older were recruited from three teaching hospitals in northern Taiwan between 2007 and 2010. Healthy controls (n = 468) were recruited from health checkups and volunteers of one hospital during the same time. Four common (5%) haplotype-tagging SNPs (htSNPs) were selected from IL6 gene to test the association between sequence variants of IL-6 and the risk of AD. Three IL-6 SNPs (rs1800796, rs1524107, and rs2066992) were associated with a reduced risk of AD adjusting for APOE e4 status, age, and sex [rs1800796: GG vs. CC, adjusted odds ratio (AOR)=0.37, 95% confidence interval (CI)=0.18-0.79; rs1524107: TC vs. TT, AOR = 0.69, 95% CI = 0.48-0.99, CC vs. TT, AOR = 0.38, 95% CI = 0.18-0.80; rs2066992: GG vs. TT, AOR = 0.39, 95% CI = 0.18-0.81]. The most common (frequency = 75%) haplotype CATT composed by four major alleles was associated with an increased risk of AD (1 copy vs. 0 copy: AOR = 2.38, 95% CI = 1.19-4.75; 2 copies vs. 0 copy: AOR = 2.95, 95% CI = 1.50-5.81). The sequence variants of IL-6 were associated with a reduced risk of AD in this Asian population." @default.
