The binding of guanosine triphosphate (GTP) and adenosine triphosphate (ATP) to oxy and deoxygenated samples of hemolysate and individual hemoglobin components (HbI and HbII) prepared from the blood of the American eel, Anguilla rostrata, was examined by membrane ultrafiltration. 2. Deoxygenated unstripped hemolysates bound significantly more total nucleoside triphosphates (NTP) than did the corresponding oxygenated solutions. The mean total phosphorus binding percentages were similar. 3. Stripped samples of hemolysate to which GTP or ATP was added at intraerythrocytic concentrations showed significant preferential binding in the deoxygenated state; samples to which the combined triphosphates were added showed no significant difference. 4. Deoxygenated triphosphate-replaced solutions of HbI and HbII did not bind significantly greater amounts of total NTP than did oxygenated solutions. 5. For all NTP-replaced samples, the differences in the mean total phosphorus binding percentages followed the pattern obtained for the binding of organic phosphates alone. 6. The three types of triphosphate additions produced binding data in disagreement with previously determined effects on hemoglobin oxygen affinity. 7. There was no significant correlation between extent of binding and increase in hemoglobin concentration. 8. The greater binding of triphosphates to HbI than to HbII may indicate functional differences in oxygen transport." @default.
