- W2000209403 abstract "Metabolism of [2-14C]thiouridine by rat liver slices was studied. 2-Thiouridine was readily converted to acid-soluble nucleotides which were identified as thio-UMP, thio-UDP, and thio-UTP. Identification of the latter two thionucleotides was based on chromatographic characteristics on anion-exchange columns and thin-layer plates as well as the nature of the products formed after partial degradation. Two thio-UDP-conjugates, thio-UDP-glucose and thio-UDP-N-acetylglucosamine, were also identified by similar methods and by the incorporation of radioactive carbohydrate. The formation of thio-UDP-glucuronic acid was also suggested. [2-14C]Thiouridine was also incorporated into rat liver RNA and the radioactivity in RNA hydrolyzates, chromatographed on anion-exchange columns and thin-layer plates, was coincidental with authentic thio-UMP. Thus, it appears that pathways exist in mammalian cells by which thiouracil can be incorporated into thionucleotides, thio-UDP-conjugates, and RNA. The similarities in the metabolites formed from uridine and thiouridine suggest that the pathways for incorporation of thiouridine into thio-UDP conjugates and RNA are identical or at least strikingly similar to those utilizing uridine." @default.
