- W2000281302 abstract "Background: Excessive alcohol consumption represents an important risk factor for the development of acute and chronic pancreatitis. Oxidative stress contributes to the development of alcoholic liver disease and appears to be involved in pancreatic inflammation as well. Aim: To investigate the effects of acute ethanol vapour administration on the antioxidant status in the liver and pancreas of adult rats by determination of enzymatic activities of glutathione peroxidase (GPX), catalase (CAT) and concentrations of reduced glutathione (GSH). Methods: Female Wistar rats weighing 230–250 g were used. Rats (n=8) were exposed to ethanol vapour in ethanol vapour chambers for 4 hours on 3 consecutive days. Control animals (n=8) were kept in ventilated control chambers for similar periods of time. Blood alcohol levels were determined immediately after treatment on the first day (mean 276±70 mg/dl) and third day (mean 255±44 mg/dl). Rats were killed under anesthesia 3 hours after termination of treatment. Pancreatic and liver specimen were collected. We determined GPX according to Lawrence and Burk (results in U/mg protein), CAT by the method of Aebi (k/g protein), and GSH as described by Tietze (nM/mg protein). Results: CAT and GPX activities decreased in the liver and pancreas with ethanol administration, but did not change significantly compared to controls (p>0.05). As demonstrated with other models of acute ethanol administration, ethanol treatment significantly decreased liver GSH content compared to controls (11.71±3.94 vs. 15.8±3.11; p<0.05). In contrast, ethanol administration did not significantly change pancreatic GSH content compared to controls (0.84±0.26 vs. 0.78±0.25, p>0.05). Conclusions: We confirm that acute ethanol vapour exposure significantly reduces GSH content in the liver which has been shown with other models as well.
