- W2000339480 abstract "Abstract 1. 1. A new method is described for the isolation of pure viable lymphocytes from 10–1000 ml of human peripheral blood. The average yield was 64%, contamination with other blood cells less than 5%. Freshly collected blood as was as blood that has been stored overnight can be used. 2. 2. Application of mechanized fluorimetric determination facilitated the analysis of lactate, pyruvate and glycerol + glycerol-1-P production and the level of ATP + ADP, glucose-1,6-P2 and 2,3-P2-glycerate in small amounts of lymphocytes. Incorporation of [32P]phosphate into acid-soluble nucleotides was also measured. 3. 3. The changes in these parameters were measured during 4-h incubations with glucose and with adenosine + pyruvate. 4. 4. Phytohaemagglutinin induced a direct decrease of the ATP +_ADP level and a direct increase in lactate and pyruvate production. After 2 h an increase was induced in the level of ATP + ADP, glucose-1,6-P2 and 2,3-P2-glycerate. At the same time glycerol + glycerol-1-P production and specific Pi incroporation were increased. 5. 5. The influence of the following inhibitors of carbohydrate metabolism on the stimulation by phytohaegmagglutinin was measured: monoiodoacetic acid, NaF, 2-deoxyglucose and KCN. In the presence of monoiodoacetic acid or NaF, phytohaemagglutinin was not able to exercise its action but in the presence of 2-deoxyglucose or KCN, phytohaemagglutinin wasa able to influence several metabolic parameters. 6. 6. The mechanism of action of 2-deoxyglucose is discussed. 7. 7. It is concluded that in vitro phytohaemagglutinin begins with the stimulation of energy consuming processes.
