These techniques include the immobilization of antibodies against the desired enzyme on such supports as Sepharose and agarose for use in low-pressure affinity chromatography (LPAC). The time required for column work can be shortened considerably by using high-pressure affinity chromatography (HPAC) instead of LPAC. The use of HPAC requires that the antibody be immobilized on supports such as silica which can withstand the high pressures associated with HPAC.
