- W2000490430 abstract "Abstract Repeated nuclear DNA sequences from Cucumis melo have been used to gather information on their modulation in cultured cells and regenerated plants as compared to the original plant material. Two types of repeated sequences have been analyzed, a 360 bp tandemly repeated sequence and two families of dispersed repeated sequences. The tandemly repeated family had previously been shown to be methylated and to exhibit a different methylation pattern in long-term callus cultures compared to hypocotyls. In this paper, we show that the methylation pattern of this sequence family is identical in short-term callus cultures and in regenerated plants with that of the starting material (cotyledons). No variation in the copy number of this sequence family has been observed whatever the tissue studied. The two dispersed repeated sequence families were found to be unmethylated but a dramatic reduction in their copy number was observed during tissue culture, this copy number being only 10% in long-term callus cultures as compared to the original plant material. Furthermore the two dispersed repeated have been identified as promiscuous DNA present both in nucleus and in mitochondria." @default.
