With copper salts, the chelate presents a characteristic ultraviolet absorbance at 270 nm, making it possible to titrate ethambutol extracted from plasma with chloroform in alkaline medium. The column which was used contained silica (LiChrosorb Si 60, 5 μm) and the mobile phase was a mixture of equal parts of water and acetonitrile, containing copper sulphate and ammonia. The internal standard was a lower homologue of ethambutol with the same chelating ability. The detection limit was 0.15 mg/l. No interference with other antitubercular agents was observed. La méthode proposée se fonde sur la capacité de l'éthambutol de former avec des cations métalliques divalents, des complexes de chélation. Celui obtenu avec les sels de cuivre, présente une absorption caractéristique à 270 nm, permettant un dosage de l'éthambutol plasmatique après extraction chloroformique en milieu alcalin. La colonne utilisée contient de la silice (LiChrosorb Si 60, 5νm) et la phase mobile consiste en un mélange d'eau-acétonitrile (50:50) contenant du sulfate de cuivre et de l'ammoniaque. L'étalon interne est un homologue inférieur de l'éthambutol présentant les mêmes propriétés complexantes. La limite de détection est de 0,15 mg/l et les autres antituberculeux employés en association ne donnent aucune interférence." @default.
