Cette revue offre un récapitulatif des antibiotiques à longue demi-vie, les aspects pharmacocinétiques et pharmacodynamiques inhérents à celle-ci, et ses principaux avantages ou inconvénients dans la gestion du traitement des infections bactériennes, à la fois pour le patient (compliance, confort) et pour la communauté (coût global, infections nosocomiales). The elimination from the blood of any drug and especially of an antibiotic is defined by its elimination half-life. The length of this half-life is then essential for the establishment of the dosing regimen in an antibacterial therapy, concomitantly with tissue diffusion, bactericidal activity, toxicity, and potential post-antibiotic effect. The length of the half-life depends on the molecule properties, protein binding, or the capture by polymorphonuclear neutrophils for transport to the site of infection. Thus, variabilities between antibiotic classes or within a specific class can occur. In this review, we summed up the major long-acting antibiotics, their pharmacodynamic and pharmacokinetic behaviors, the advantages and disadvantages in the antibacterial chemotherapy, at once for the patient (compliance, comfort) and for the community (treatment costs, emergence of resistant pathogens)." @default.
