In vitro studies have been carried out to determine the effects of cortisol 21-phosphate on the template efficiency of rat-liver chromatin as measured by its ability to incorporate labelled UMP into RNA in the presence of transcriptase from Micrococcus lysodeikticus 2. These experiments show that cortisol increases the template capacity of chromatin by 10–12 % and that this increase is dependent upon the hormone concentration. A concentration of 4 μg cortisol per 0.5 ml reaction mixture has the maximum stimulatory effect. 3. The increased efficiency of priming is not due to the stimulation of the exogenous transcriptase by cortisol, in that the template capacity of calf-thymus DNA was not increased by cortisol. 4. These findings have been discussed along with some possible mechanisms of cortisol action. It is concluded that cortisol acts at the chromatin level by increasing the capacity of chromatin to prime for RNA synthesis." @default.
