- W2000820400 abstract "The brush cell is a cell type of the respiratory epithelium that is characterized by an apical tuff of microvilli protruding into the lumen. It expresses proteins of the taste transduction pathway and, hence, is considered to serve chemosensory function. The expression of effector molecules that confer signalling to neighbouring cells/nerve fibres, have not yet been identified however. Here, we investigated the possibility of acetylcholine (ACh) being produced by brush cells in the mouse trachea utilizing immunohistochemistry and a BAC transgenic mouse strain expressing eGFP under control of the promoter of the ACh synthesizing enzyme, choline acetyltransferase (ChAT) [1]. Brush cells were identified by immunoreactivity to villin. Double-labelling immunohistochemistry in wild-type mice revealed that villin-immunoreactive cells are also labelled by antisera against ChAT and VAChT (vesicular ACh transporter). Whole-mount preparations of tracheas from ChAT(BAC)-eGFP mice displayed 6242 (mean of 6 animals) solitary eGFP expressing cells scattered along the entire length of the trachea, about two thirds of them located in epithelial stretches overlaying cartilage-free areas. These cells were immunoreactive for VAChT and villin but not for PGP9.5, a marker of neuroendocrine cells. Immuno-electron microscopy utilizing an anti-GFP antibody labelled immunoreactive epithelial cells, which were identified as brush cells by their characteristic ultrastructure. Confocal laser scanning microscopy of PGP9.5-immunolabelled sections revealed that a subpopulation of cholinergic brush cells, but not all of them, establishes contacts with presumably sensory nerve fibres. These data demonstrate the expression of two key proteins of cholinergic signalling, i.e. the ACh synthesizing enzyme ChAT and the transporter responsible for translocation of ACh into vesicles, VAChT, by tracheal brush cells." @default.
