- W2000914141 abstract "The pathways of de novo lipid synthesis from precursor glycerol and acetate were examined in mammalian reticulocytes. 70% of precursor glycerol is incorporated into the 3-carbon backbone of phospholipids, mainly phosphatidylcholine and phosphatidylethanolamine. There is little labeling of sphingomyelin and phosphatidylserine. Approximately equal amounts of the remaining 30% of precursor glycerol are incorporated into fatty acids and cholesterol after shunting through the Emden-Meyerhof pathway. The amount of glycerol incorporated into reticulocyte membrane phospholipids is only 10 pg/ml of cells per h at 37 °C, representing less than 0.1% of the total membrane phospholipid content. Although the majority of precursor acetate is incorporated into neutral lipids, a significant amount of acetate is incorporated into the 3-carbon glycerol backbone of phospholipids via the Emden-Meyerhof pathway. Incorporation of both acetate and glycerol into reticulocyte lipids is temperature dependent and requires energy, and does not take place in mature erythrocytes. Reticulocytes can synthesize both cholesterol and phospholipids de novo from either acetate or glycerol, utilizing interlocking pathways which are common to other mammalian cells." @default.
