- W200092703 abstract "Multicomponent complexes of miconazole with different acids and cyclodextrins The miconazole (Mi) is an imidazole antifungal agent with a low aqueous solubility (<0.4mg.ml -1 ). Mi is a weak base showing a higher solubility at low pH values. The purpose of the present study is to investigate the combined effects of different acids and cyclodextrins (CDs) on the aqueoous solubility of Mi. Phase solubility studies have shown that the solubility of Mi increase with the β-CD and hydroxyprophy-β-CD. (HP-β-CD) concentration (A p diagrams type according to Higuchi and Connors). Binding constants (Kc) were found to be equal to 1.8 x 10 6 and 1.4 x 10 6 M -1 respectively with β-CD and HP-β-CD. Mi's solubility in a 15 mM β-CD solution was 140 times higher than the solubility in water. With a 50 mM HP-β-CD a 200-fold increase was obtained. Below pH 4, -significant differences in solubilities were observed depending on the nature of the buffer solution. In pH 3 phosphate, citrate, lactate, acetate and gluconate buffer solutions, the solubility of Mi was 783, 107, 864, 622 and 1359 μg/ml respectively. The combined use of acids and HP-β-CD allows to obtain multicomponent complexes giving a solubility higher than 10 mg of Mi per ml. These results show that it would be possible to formulate a solution of Mi for intravenous use without surfatant." @default.
