- W2002068002 abstract "Experiments were performed to study the inhibitory effects on gonadotropin release of intrahypothalamic implants of gonadal steroids with particular reference to the possibility that their primary effect is at the pituitary level (via steroid transport through the hypophyseal portal vessels). Crystalline dihydrotestosterone propionate (DHTP, 200 µg, estradiol benzoate (EB, 10 µg), cortisol acetate (200 µg), progesterone (400 µg), and cholesterol were implanted in the medial basal hypothalamus of castrated male and female rats. At different times after implantation, basal plasma LH levels were determined, while changes in pituitary responsiveness were monitored by measuring the plasma LH response to iv injections of synthetic LRF. At the end of the experiment (1 week), plasma LH was suppressed by DHTP, EB and proges-terone, in that order of effectiveness. No significant change in basal plasma LH or LRF response was found at any time in animals implanted with cortisol. The LH-suppressing effect of DHTP implants was paralleled by a progressive decline in the response of the pituitary to LRF, but no changes n i pituitary sensitivity accompanied the plasma LH inhibition found with progesterone. These findings applied equally to male and female rats. EB implants (10 µg) suppressed plasma LH to a level approaching that found with DHTP (200 µg). After an initial drop in LRF response, significant only in the female, pituitary sensitivity returned to normal n i EB-implanted males, and in females, increased above the levels found in controls on days 1, 3 and 7. We interpret these experiments to indicate that inhibitory (negative) feedback effects of estrogen and progesterone are exerted at the hypothalamic level, and stimulatory (positive) feedback effects of estrogen at the pituitary level. DHTP may suppress LH secretion by direct action on the pituitary, and/or at the hypothalamic level, with the pituitary showing decreased responsiveness secondary to removal of endogenous LRF.
