- W2002956171 abstract "Glucose, by inhibiting the outward transport of calcium across the cell membrane, and theophylline, by causing an intracellular translocation of calcium, are both thought to increase the concentration of calcium in the cytosol of the beta cell. In support of this concept, it is here shown that theophylline increases the efflux of 45Ca from islets perifused in the presence or absence of extracellular calcium, probably by enriching the pool of cellular calcium available for outward transport. Glucose markedly reduces the effect of theophylline on the 45Ca efflux, suggesting that the sugar prevents the cationic outflow otherwise associated with theophylline-induced calcium redistribution. Epinephrine facilitates the outward transport of calcium, both in the absence and presence of glucose. The epinephrine-induced increase in the 45Ca efflux is a transient phenomenon, as if epinephrine were to simultaneously deplete the pool of calcium available for outward transport. Such a depletion could be due to an intracellular redistribution of calcium opposite to that ascribed to theophylline, since epinephrine also antagonizes the effect of theophylline on the 45Ca efflux. It is hypothesized that the ubiquitous inhibitory effect of epinephrine on insulin release is due to a glucose-independent facilitation of both the outward transport and vacuolar uptake of calcium." @default.
