- W2003245606 abstract "In view of great species differences in biology of polymorphonuclear cells, and nonavailability of basic data on buffalo PMN cells for assessing their functional activity, the present work on the immuno-defence system involving protein synthesis and O − 2 production was undertaken to highlight the immunomodulatory role of thyroxine. Digitonin, LPS and Con-A activation generated superoxide, which was monitored by NBT reduction. The study suggested that concanvalin A (Con-A) and T 4 were able to synergetically increase the production of superoxide and H 2 O 2 . The likely involvement of thyroxine in activation was studied by [ 125 I]thyroxine incorporation, which was significantly increased due to activation. In contrast, aflatoxin B 1 together with Con-A caused a significant decrease ( P < 0.05) in incorporation of [ 125 I]T 4 . Optimum time dependence in [ 14 C]leucine incorporation by buffalo PMN cells was found to be 30 min and the factors like T 4 (7.7 ng/ml) and glutathione (400 μg/ml) significantly enhanced the incorporation. In contrast, antiinflammatory agent, indomethacin (40 μg/ml) inhibited protein synthesis in PMN cells; while puramycin also significantly lowered the [ 14 C]leucine incorporation. Total [ 14 C]leucine incorporation in acid extractable cationic proteins and peptides, known for their antibacterial properties was found to be 30–40% when separated on AU-PAGE. The studies revealed the in vitro immunomodulatory role of T 4 in O − 2 , H 2 O 2 production and cationic protein synthesis by the activated PMN cells of buffalos. En raison des grandes différences dues à l'espèce dans la biologie des cellules polynucléaires et de l'absence de données de base concernant l'activité fonctionnelle des polynucléaires du buffle, le présent travail a été entrepris pour éclaircir le rôle immunomodulateur de la thyroxine dans le système immunitaire de défense impliquant la synthèse des protéines et la production d'O 2 . L'activation par la Digitonine, le LPS et la Con-A génère du superoxide mis en évidence par réduction du NRT. L'étude suggère que la Con-A et le T 4 sont capables d'augmenter de façon synergique la production de superoxide et de H 2 O 2 . L'implication probable de la thyroxide dans l'activation a été étudiée par incorporation de thyroxine (125) dont le taux a été significativement augmenté lors de l'activation. En revanche, l'aflatoxine B 1 avec la Con-A provoque une diminution significative ( P < 0.05) de l'incorporation de T 4 (125). Le temps de dépendance optimal de l'incorporation de leucine C 14 par les polynucléaires de buffles était de 30 min et les facteurs T 4 (7,7 μg/ml et glutathione (400 μg/ml) ont augmenté significativement l'incorporation. En revanche, l'indomethacin anti-inflammatoire utilisée à dose de 40 μg/ml inhibe la synthèse des protéines dans les polynucléaires. La puramycine diminue également l'incorporation de leucine C 14 . L'incorporation de leucine C 14 dans les protéines et peptides connues pour leur activités antibactériennes étaient de 30–40% après séparation sur AU-PAGE. L'étude révèle le rôle immunomodulateur de T 4 sur la production de O 2 et de H 2 O 2 et sur la synthèse des protéines dans les polynucléaires activés du buffle." @default.
