Novinski, 1974, Virology 60, 595–601), have been purified from [3H]arginine- and [35S]- and [3H]methionine-labeled virus on an agarose column in 6 M guanidine-HCl. The gp 35 aggregates under these conditions and elutes with the flow through of the column; it was therefore further purified by sodium dodecyl sulfate gel electrophoresis. After removal of terminal sialic acid residues the two glycoproteins were oxidized and digested with trypsin, and the resulting peptides were separated on a cation-exchange column. Comparison of the peptide maps of gp 85 and gp 35 suggests no major sequence relationship between the two glycoproteins. A protein which migrates more slowly than gp 85 on a sodium dodecyl sulfate-polyacrylamide gel (approximately 92,000 daltons) can be immunoprecipitated from pulse-labeled infected chicken embryo fibroblasts using an anti-gp 85 serum. Comparison of the [3H]arginine-labeled peptides of this protein with the peptides obtained from gp 85 and gp 35 shows that the 92,000-dalton molecule contains the peptides of both viral glycoproteins and can therefore be considered their common precursor (pr 92gp according to proposed nomenclature, T. August, personal communication). The arrangement of the two envelope glycoproteins on the common precursor has been established. The gp 85 is located in the N-terminal position, and gp 35 is in the C-terminal position." @default.
