- W2004103658 abstract "SUMMARY The anti-arrhythmic activity of dehydrobenzperidol (DBP) was studied during halothane anaesthesia in dogs. Arrhythmias were provoked by single intravenous injections of catecholamines. Two groups of dogs were studied. One received norepinephrine and the odier epinephrine. The dose just sufficient to initiate arrirythmias was defined as the “threshold”. Electrocardiogram, blood pressure and heart rate were monitored. The principal findings were:— 1 The dog heart was “sensitized” to epinephrine and norepinephrine by halothane. 2 DBP did have an anti-arrhydimic action. A dose of 0.150 mg/kg roughly doubled the “threshold”. 3 DBP had significant alpha-adrenergic blocking properties. Beta-blocking properties were lacking. 4 The anti-arrhythmic effect of DBP was due to the hypotension mediated by die alpha-adrenergic blockade. This indirect modification of the response to catecholamines may be specific for DBP. ZUSAMMENFASSUNG An Hunden wurde während Halodiannarkosen die anti-arrhythmische Wirkung von Dehydrobenzperidol (DBP) untersucht. Die Arrhythmien wurden mit i.v. Einzeldosen von Katecholaminen provoziert. Es wurden zwei Gruppen von Hunden untersucht: Eine erhielt Noradrenalin, die andere Adrenalin. Die Dosis, die gerade ausreichte, um Arrhythmien hervorzurufen, wurde als »Schwelldosis« bezeichnet. EKG, Blutdruck und Herzfrequenz wurden apparativ überwacht. Die wesentlichsten Ergebnisse waren: 1 Das Hundeherz wird durch Halothan auf Noradrenalin und Adrenalin sensibilisiert. 2 DBP ist antiarrhythmisch wirksam: 0,15 mg/kg Körpergewicht erhöhten die »Schwelldosis« auf etwa das Doppelte. 3 DBP hat signifikante alpha-adrenergische Blockereigenschaften. Beta-blocker-Eigenschaften fehlen hingegen. 4 Die anti-arrhythmische Wirkung von DBP erklärt sich aus der, durch die alpha-adrenergische Blockade hervorgerufenen, Hypotonie. Diese indirekte Modifikation der Reaktion auf Katecholamine scheint spezifisch für DBP zu sein." @default.
