- W2005057614 abstract "The application of the arbitrarily primed PCR (AP-PCR) procedure to generate randomly amplified polymorphic DNA (RAPD) fingerprints for the study of the taxon Porphyromonas gingivalis was investigated. Nine human strains and seven animal strains of P. gingivalis as well as eighteen strains other than P. gingivalis were analysed. Four nanomer primers of random sequence were evaluated for their ability to distinguish genetic diversity. Three primers generated RAPD fingerprints that allowed the sixteen strains to be differentiated; two of the primers yielded species-specific markers, and two of the primers permitted biotype distinction. Cluster analysis of the RAPD fingerprints revealed two major phenetic groups that matched the human and animal biotypes. Our results indicate that AP-PCR (i) can generate strain-specific fingerprints, (ii) confirms genetic heterogeneity and the biotype grouping of the P. gingivalis taxon, and (iii) enables identification of potential genetic markers at the species, biotype and subtype levels and is thus a promising tool for bacterial systematics. Our results also underline the potential of AP-PCR for epidemiological studies of periodontal pathogens. Nous avons appliqué à l'étude du taxon Prophyromonas gingivalis l'utilisation des empreintes RAPD obtenues par la PCR avec amorce arbitraire (AP-PCR). Pour cette étude, 9 souches de P. gingivalis d'origine humaine, 7 souches d'origine animale, et 18 souches autres que P. gingivalis ont été retenues. Quatre amorces nanomères de séquence aléatoire ont été évaluées dans leur aptitude à mettre en évidence la diversité génétique. Trois amorces ont produit des empreintes RAPD permettant la différenciation des 16 souches; deux amorces ont permis d'identifier des marqueurs génétiques de l'espèce, et deux permettaient de reconnaîre deux biotypes. Une analyse en grappes des emprientes RAPD a révélé l'existence de deux groupes phénétiques majeurs qui correspondaient aux biotypes humain et animal. Nos résultats indiquent que l'AP-PCR (i) peut produire une empreinte spécifique de chaque souche, (ii) confirme l'hétérogénéité génétique et le groupement en biotypes du taxon, et (iii) permet d'identifier des marqueurs génétiques potentiels au niveau de l'espèce, du biotype et du sous-type, le révélant ainsi comme un outil prometteur en systématique bactérienne. Cette étude révéle en outre que l'AP-PCR offre un potentiel d'application dans l'épidémiologie des pathogènes du parodonte." @default.
