- W2005263314 abstract "Hydrogen sulfide (H2S) is signaling molecule, vasodilator, cytoprotector, antioxidant and it may be involved in pathogenesis of age-associated heart diseases. The question remains open about H2S synthesis and utilization in myocardium within aging what became our study objective. 60 male Wistar rats of three age groups (1–2 months, 6–8 months, 24–26 months) were randomized into subgroups: control (normal saline), NaHS (3 mg/kg i.p.), DL-propargylglycine (50 mg/kg i.p). Modulators of H2S metabolism were administered for 14 days. It was determined the level of free and acid-labile H2S (Wiliński B., 2011), cystathionine gamma-lyase (CSE), cysteine aminotransferase + 3-mercaptopyruvate sulfurtransferase (CAT/3MST), sulfite oxidase, thioredoxin reductase activity in myocardium. H2S level, CSE, CAT/3MST, sulfite oxidase, thioredoxin reductase activity in myocardium of 24–26-month-old rats were 30–50% lower than in myocardium of 6–8 and 1–2 month-old rats. Propargylglycine administration caused decrease in H2S level and CSE activity on 33.3 and 78.3% in 24–26 month-old rats, 22 and 61.8 % in 6–8 month-old rats, 14 and 50.3% in 1–2 month-old rats (p < 0.05). NaHS administration resulted in increase of H2S level on 52.6% in myocardium of 24–26 month-old rats, 16.5% in 6–8 month-old, 10.5% - 1–2 month-old and did not affect CSE and CAT/3MST activity. Propargylglycine administration caused decrease but NaHS caused increase in thioredoxin reductase and sulfite oxidase activity only in 24–26 month-old rats. Thus the balance between H2S synthesis, utilization and storage in myocardium is affected in aging and role of CSE/H2S pathway is increasing." @default.
