- W2005440045 abstract "In DNA-RNA hybridization reactions using pulse-labeled whole RNAs from the ciliated protozoan, Tetrahymena pyriformis, the presence of two general classes of reactants, a highly redundant fast component and a less redundant slow component, is indicated by saturation, kinetic and competition experiments. RNA stocks enriched for labeled rRNA by the combined techniques of long-pulse labeling and sucrose gradient fractionation show that the fast component is associated with the ribosomal region; the slow component is associated with short-pulse labeling and the more rapidly sedimentable fractions. The role of the fast component is further demonstrated in kinetic and competition experiments made with stocks artificially constructed of the two isolated components. The fast component saturates at low concentration and in kinetic plots early, is efficacious in pre-annealing, and contributes to low concentration asymptotes and cross-overs in competition hybridization experiments. The presence of such heterologous components to a greater or lesser degree contributes to discernible subtleties in whole complex RNA-DNA competition hybridization curves, often neglected but evidently present in earlier studies." @default.
