- W2005753951 abstract "Chemical peeling is one of the dermatological treatments available for certain cutaneous diseases or aesthetic improvement; it consists of the application of one or more chemical exfoliating agents to the skin. However, the scientific background and adequate technical specifications for this procedure have not yet been established. Among the many chemical peeling agents, TCA and phenol have been used for the treatment of several skin diseases. Both agents have tissue injury effects through the papillary dermis to the upper reticular dermis. The aim of our present study was to examine the distribution pattern of P-cadherin and E-cadherin in peeled skin, because both cadherins contribute to the normal architecture of the skin, and aberrant suprabasal P-cadherin expression has been found in acanthotic skin. Murine monoclonal antibodies (mAbs) against HECD-1 (human E-cadherin), NCC-CAD-299 (human P-cadherin) and DE-K10 (keratin 10; from Dakopatts, Glostnep, Denmark) were used on cryostat sections. Skin specimens were obtained from the upper arm of three healthy adults. First, phenol (100%), 40% TCA or 60% TCA in water were applied for adequate times, and the peeled sites were semi-closed by standard methods. Two days or seven days after peeling, punch biopsies were performed and the skin specimens were prepared for hematoxylin eosin (HE) staining and immunohistochemical studies. P-cadherin expression was absent or very weak on the basal cell surfaces by day 2. Seven days after peeling, P-cadherin was clearly distributed in a continuous granular pattern over the cell surface of the entire epidermis of the 40% TCA treated skin, in a weak granular pattern on a few suprabasal cells and basal cells of the phenol treated skin, and very weakly expressed on the lateral surfaces and in the cytoplasm of basal cells of the 60% TCA treated skin. Based on the present results and previous reports, it is likely that there are distinct patterns of P cadherin expression. Furthermore, a specific type of P cadherin expression might be involved in wound healing in general, which could provide new insights into tissue repair mechanisms after chemical peeling." @default.
