- W2006499613 abstract "A cell-free extract from mouse L cells directed by 26 S RNA from Semliki Forest virus-infected cells, which is capable of synthesizing both the core and the envelope proteins of the virus, incorporates label from formyl-[35S]methionyl-transfer RNAf only into core protein. Peptide maps of tryptic digests of the complete incorporation mixture labelled with this precursor show only a single labelled peptide. In extracts in which elongation is inhibited by diphtheria toxin and NAD, only one dipeptide, methionyl-asparagine, can be identified. When virus-infected cells are released from hypertonic inhibition of initiation of protein synthesis, only capsid protein is labelled at very early times after resumption of protein synthesis. If labelling is carried out immediately after exposure of the cells to hypertonic medium, the synthesis of the core protein is more rapidly depressed than that of the other virus-specific proteins. This evidence shows that a single initiation site is used for the synthesis of the structural proteins of Semliki Forest virus, both in vivo and in vitro. It appears that the post-translational cleavage system which generates the viral proteins operates with great efficiency in this cell-free system." @default.
