- W2006703072 abstract "The uptake of H3-proline, in vivo and in vitro, was used to assess the rate and amount of collagen synthesis in femoral bone of rats and guinea pigs. Microradioautographic analysis was employed to measure in vivo cellular uptake of radioproline. Although ascorbic acid deficiency and large doses of parathyroid extract caused marked inhibition of cellular incorporation of the isotope, variations in endogenous parathyroid secretion did not significantly influence this incorporation. Similarly, the in vitro synthesis of rat femoral collagen was clearly inhibited by parathyroid extract, while parathyroidectomy produced no detectable alteration in collagen formation. However, varying media calcium concentrations did significantly influence the in vitro synthesis of collagen.It is suggested that pharmacological doses of extract may not be accurately regarded as an exaggeration of physiological parathyroid hormone actions, and that the parathyroid control of calcium homeostasis is not appreciably mediated by any immediate effect on bone-forming (osteoblastic) processes. Differences observed between the two areas of femoral bone studied (metaphyseal and diaphyseal areas) are also noted." @default.
