- W2007059499 abstract "OBJECTIVE: Trophectoderm (TE) biopsy can be performed relative to blastocyst growth on either D5 or D6 of embryo development. The aim of this study was to examine the timing of TE biopsy relative to chromosome composition and frozen blastocyst transfer (FBT). DESIGN: Retrospective study. MATERIALS AND METHODS: Seventy patients (mean maternal age = 37.4yrs) were recruited for TE biopsy with comprehensive chromosomal screening (CCS) followed by vitrification with a subsequent FBT. Blastocysts were classified for biopsy when a distinct inner cell mass was identifiable either on D5 (n=255) or D6 (n=180) of embryo development. CCS was performed by comparative genomic hybridization (CGH) (Reprogenetics) or single nucleotide polymorphism (SNP) based microarray (RMA-NJ). RESULTS: D6 biopsied blastocysts were considered morphologically slow in development on D5. However, there was no direct correlation between D5 morphology (compacting, morula or early blastocyst) and D6 aneupoidy. Similarities were observed in the number of euploid D5 and D6 blastocysts (D5=52.1% v. D6=47.8%; ns). The most frequent chromosomes involved in aneuploidy on both D5 and D6, contributed to over 50% of autosomal errors (15, 16, 19, 21 & 22). Sex chromosome aneuploidies (D5=11.8% v. D6=17.6%; ns) and chaotic blastocysts (involving ≥3 errors) (D5=10.5% v. D6=10.2%; ns) were comparable across the days of biopsy. Initial clinical data following FBTs with only D5 euploid blastocysts (n=34) or only D6 euploid blastocysts (n=16) indicate a trend towards a better outcome following D5 biopsy (clinical pregnancy rate D5=91% v. D6=69%; implantation fetal heart rate D5=73% v. D6=57%; both ns). The CCS technique employed had no bearing on results or outcome when analyzed separately. CONCLUSIONS: Analysis of the chromosome composition of human D5 and D6 blastocysts revealed similarities in relation to type and incidence of aneuploidy. Preliminary outcome data indicate a trend towards an advantage following biopsy of human blastocysts on D5 of development." @default.
