- W2007132322 abstract "Neuroticism is an enduring, heritable personality trait and a major risk factor for neuropsychiatric illness. Therefore, it should have a neurophysiological basis and be reflected in physiological measures. A strong magnetic pulse delivered to motor cortex produces motor evoked potentials (MEPs) in contralateral muscles. MEP amplitude is reduced when a weak conditioning pulse precedes such a stimulus by 2–5 ms. The conditioning pulse is thought to activate inhibitory GABAergic interneurons. Neuroticism is high in OCD where patients have abnormally low levels of intracortical inhibition. In a pilot study intracortical inhibition and neuroticism were inversely correlated in OCD patients and controls. Here, we studied the relationship between intracortical inhibition and NEO personality inventory neuroticism and PANAS negative emotionality in 20 male, non-patient subjects. TMS was delivered singly and in pairs 3, 4, 10 and 15 ms apart. The degree of inhibition produced by the conditioning pulse was expressed as the ratio of mean MEP amplitude from single pulses to that from pairs with each interval. There were significant negative correlations between intracortical inhibition at an interstimulus interval of 3 ms and both NEO neuroticism (p = 0.02, r = 0.49) and PANAS negative affect (p = 0.0048, r = 0.607). No other personality domain was significantly correlated with intracortical inhibition. These findings confirm our impression that negative emotionality may be influenced by the balance of cortical inhibition and excitation. The data suggest that lowered intracortical inhibition might be a promising intermediate phenotype in studies of brain mechanisms influencing a personality trait central to psychopathology." @default.
