Using [5 -3 H]-uracil incorporation into acid-insoluble material or 14 CO 2 release from [2 -14 C]-uracil, distinct lag times in the labeling of both products were determined by six different types of tracer kinetics as a measure of the metabolic pool size. This size was calculated to be 11 pmoles/10 7 cells in heterotrophically grown cells. The metabolically active pool of uracil which increased by preloading with uracil in a time-dependent reversible and saturable process seems to be of functional significance during high rate of RNA synthesis. Its size decreased to almost zero during light-induced RNA synthesis, was shown to be growth-phase dependent to different cell types and apoplastic mutants and was undetectable in autotrophically grown cells." @default.
