• W2007704417 abstract "The osmotic contribution of glutaraldehyde and that of the fixative vehicle in the fixation of the rat lung has been studied by transmission and scanning electron microscopy and by morphometry. Hypo-, iso-, and hypertonic fixatives (osmolarity ranges for total solution, 145–1080 m Osm ; buffer, 40–920 m Osm ; glutaraldehyde, 95–495 m Osm (1–5%)) have been instilled through the trachea. The total osmolarity was maintained during subsequent rinsing and postfixation. Hypotonic fixatives led to spherical deformation of the erythrocytes (EC) and swelling of the air-blood tissue barrier. Isotonic fixatives (300–360 m Osm ) preserved the biconcave discoid shape of EC. Hypertonic fixatives induced shrinkage of the cytoplasm and extracted plasma water from the alveolar capillaries; as a result, plasma gelled and the air-blood tissue barrier collapsed onto EC. When the concentration of glutaraldehyde was increased from 1 to 5% in 0.05 M sodium cacodylate buffer, the air-blood tissue barrier became thinner. It is concluded that both the percentage of glutaraldehyde and the molarity of the vehicle in the fixative are of importance for the preservation of lung tissue. A formula is derived which predicts the rate of dimensional changes as glutaraldehyde and total fixative osmolarity are varied." @default.
