- W2007704616 abstract "Abstract The time course of viral RNA synthesis, virus maturation, and cell surface alteration was studied in a clonal line of mouse 3T6 cells infected and transformed by the Harvey strain of the murine sarcoma-leukemia virus complex, H-MSV(MLV). The majority of cells (80%) were infected and gave rise to transformed foci after exposure to 2–3 focus-forming units of virus/cell for 60 min. Most cells became spindle shaped by 24 hr, and virtually all cells assumed a round shape, presumably the final stage of morphological transformation, within 3–4 days. The transcription of virus-specific RNA, as measured by hybridization of RNA from infected cells with the [ 3 H]DNA product of the virion DNA polymerase, was detected at 7 hr after infection, and reached maximal levels at 25 hr. Virus replication in newly infected cells was measured by the appearance in the culture medium of (1) [ 3 H]uridine labeled particles banding at a density of 1.14–1.16 g/ml in sucrose density gradients, (2) particles with RNA and DNA-directed DNA polymerase activity banding at a similar density, and (3) focus-forming units. Of the three methods, the most sensitive was the assay for polymerase, which detected the formation of extracellular virus at 14–15 hr after infection. Maximal virus production occurred at about 63 hr. Surface alterations, as revealed by agglutination of cells by concanavalin A, were detected 22 hr after infection and reached maximal levels at 63 hr." @default.
