- W2007760845 abstract "In both humans with Alzheimer's disease (AD) and AD mouse models, cerebral blood flow is decreased by as much as 30% relative to age-matched controls. Although the cause of this blood flow deficit remains unclear, it could have cognitive consequences and may decrease the clearance of amyloid-beta, thus accelerating AD progression. We examined the role of microvascular factors in this brain blood flow decrease. Using two-photon excited fluorescence microscopy, we imaged the cortical vasculature in transgenic AD (B6. Cg-Tg(APPswe, PSEN1dE9)85Dbo/J) and wildtype mice with cranial windows. We labeled blood plasma with Texas Red-dextran and leukocytes and platelets with rhodamine 6G. We also added Hoechst to label nuclei, allowing us to distinguish leukocytes. Individual capillaries were scored as flowing (circled; Fig. 1a) or stalled (arrow; Fig. 1a) based on whether motion of unlabeled erythrocytes was observed over a ∼2 s observation time. In AD mice (12 mice, 58 +/-17 (mean+/- standard deviation) weeks old), we found the fraction of capillaries with spontaneously stalled blood flow was 1.6% +/ 0.7%, four times higher (0.4% +/- 0.3%) than in age-matched wildtype littermates (9 mice, 59 +/- 21 weeks old, Fig. 1b). No flow obstructions were observed in arterioles or venules. In the AD animals, about two-thirds of the stalls were caused by a leukocyte that plugged the capillary segment. In repeated imaging of the same vessels, we found that most of the stalled capillaries reperfused within 2 min., but 20% remained plugged at all imaged time points (∼15 minute intervals) out to an hour. We also found that 30% of the reperfused capillaries were stalled again at one of the later imaging times. Finally, we observed some capillaries were alternately flowing and stalled on successive imaging days over several weeks (Fig. 1c)." @default.
