- W2007806939 abstract "Immunophenotyping by multiparameter flow cytometry can provide a rapid, accurate method for both identifying unique cell populations and describing their functional status. This chapter discusses the role of intermediate filament (IF) and multiparameter flow cytometry in the study of solid tumors. It also discusses the principle and technique of flow cytometry, and the procedures of immunohistochemistry and multiparameter flow cytometry. The combination and the use of immunochemical labeling of the IFs and multiparameter DNA flow cytometric analysis can help to separate the data related to specific immunophenotypes, to collect data from a variety of different classes in the same run, and to relate these specifically to the population of interest from those in the total population. With this combination, it is virtually possible to visualize the presence and distribution of multiple cell characteristics directly and make calculations with them in relation to each other. A major advantage of applying multiparameter flow cytometry on routinely processed formalin-fixed, paraffin-embedded specimens is that morphological information is still available; however, important issues in implementing multiparameter DNA flow cytometry are a solid quality control and management system that have to be set up to guarantee that the data obtained are robust and reliable. Multiparameter flow cytometry completed with advanced sorting facilities might become a serious and economically favorable competitor of other more laborious methods, such as laser capturing or automated image analysis, where only the image is retained and not the actual cell." @default.
