- W2008080471 abstract "The buoyant densities of DNA isolated from intact monkey pox virus (MPV) and of DNA isolated from the cytoplasmic fraction of MPV-infected CV-1 cells 4 h post infection were determined by CsCl density gradient centrifugation to have an identical value of 1.69 g/cm3. Uninfected cell DNA analyzed in a methylated albumin kieselguhr column eluted in a single peak from the column with 0.67 M saline buffer whereas MPV-infected cell DNA eluted in two peaks; one with 0.67 M and one with 0.71 M saline buffer. The 0.71 M peak was approximately 60 % of the quantity of the cellular DNA. MPV heated at 100 °C for 10 min exhibited a 32.5 % increase in absorbance and a 34.6 % increase when heated in presence of 1 % formaldehyde; treatment with 1 % formaldehyde alone caused no increase in absorbance; 50 μg/ml deoxyribonuclease caused a 37.5 % increase in absorbance. MPV DNA was shown to possess a Tm of 84.8 °C, corresponding to a G+C content of 37.8 %. A molecular weight of 159 · 106 was estimated by co-sedimentation with T2 DNA." @default.
